Instrument description:
The MARS6 automatic extractor (CEM) allows for the automated extraction of lipids from various matrices (soils, sediments, peats, loess, plants). The Multivapor evaporator (Büchi) allows for the simultaneous evaporation of extraction solvents from 12 samples.
Principle of the analysis:
Lipid extraction from the samples is performed in glass tubes inserted into Teflon reactors, with a magnetic stirrer ensuring sample homogenization. The dried and ground sample is placed in the glass tube with a solvent or solvent mixture (most often DCM:MeOH 1:1 v/v). Microwaves allow the sample temperature to rise to 100 °C. After extraction, the sample returns to room temperature and atmospheric pressure. The total extract is filtered before the solvent is evaporated using a Multivapor (Büchi). The total extract is then placed in a new glass tube and placed in a water bath. Under the combined effect of heat and vacuum, the solvent is evaporated and condensed on a coil cooled by a chiller. The solvent is collected in a flask.
